Our laboratory is interested in understanding the molecular and cellular signaling mechanisms that underlie critical pathological processes such as cancer, obesity and inflammation, with special attention to asthma. Specifically, we are studying the importance and mechanisms of action of a number of key proteins in these processes such as the atypical PKCs and their regulators Par-4 and p62. The latter is very important since we have demonstrated that its genetic ablation in mice lead to obesity suggesting a role of p62 in the control of the differentiation of fat cells, also known as adipogenesis. This implication of p62 in the control of cell differentiation may also be critical for its role in the control of Th2 differentiation and asthma as well as in cancer. In this regard, the other atypical PKC regulator, Par-4, is a pro-apoptotic protein whose genetic inactivation gives rise to hyperinflammation in vivo, and higher sensitivity to tumor development, particularly in the prostate. The immediate goals of our laboratory are to elucidate the precise mechanisms of action of the atypical PKC-p62 and Par-4 complexes in cell growth and differentiation in cancer, obesity and asthma. For this we use molecular and cellular systems, as well as knock out mice that serve to validate in vivo the importance of our findings and to discover potentially novel and unanticipated functions of these protein signaling regulators.
